200754800 - The use of cDNA microarrays to develop biomarkers of environmental stress in salmonids and other fishes

Sponsor: US Geological Survey - Cook

Short description: This project will use cDNA microarrays to identify molecular biomarkers of environmental stress, such as exposure to high temperatures or toxicants, in fish. Our goal is to develop a tool for use in ecophysiological monitoring, research, and management.

Location: Mainstem on the ground/Multiprovince province, Systemwide subbasin

Budgets: FY08: $284,065 | FY09: $288,287

Primary species:
Anadromous: All Anadromous Salmonids
Anadromous: Pacific Lamprey
Resident: Pacific Lamprey

Objectives

ObjectiveDescriptionSubbasin planStrategies
Develop a lamprey-specific cDNA microarray We will create a cDNA microarray to characterize the stress responses of lampreys at the genome level Deschutes An example from a relevant subbasin (other examples exist): Risks to lamprey populations include stream habitat degradation (e.g., increased water temperatures and poor riparian areas)
Expose salmonids and lampreys to stressors We will expose juvenile salmonids and adult lampreys to common environmental stressors associated with things such as global warming and habitat degradation. Included in this would be exposure to thermal insults and contaminants. MA: (1) Meet state and federal water quality standards under the Clean Water Act; (2)Implement actions to reduce toxic contaminants in the water; (3)maintain water temperatures within the tolerance range of native fish species.
Identify reliable biomarkers for basin fishes Using a cDNA microarray, we will identify genes and their products in salmonids and lampreys that respond significantly to key environmental stressors and could serve as reliable biomarkers MA: (1) Meet state and federal water quality standards under the Clean Water Act; (2)Implement actions to reduce toxic contaminants in the water; (3)maintain water temperatures within the tolerance range of native fish species.
